The JLA has faced many villains in the comics but these 10 were the very first that they encountered.

The Justice League formed when the greatest superheroes from DC Comics had to team up to defeat a threat that was too powerful for just one hero to tackle alone. The team officially started in The Brave and the Bold, but they quickly moved into their own comic book series. Since then, the team has remained the cornerstone of DC Comics.
However, while the team started in 1960 in The Brave and the Bold #28 by Gardner Fox and Mike Sekowsky, that was not actually the first time they teamed. Later in Justice Leauge, the story of their first team-up was told, and then even later, a missing story was revealed that proved that they fought some major villains before their comic book started.
10 White Martians
The first time the Justice League teamed up to work together was before they became an official team. Green Arrow uncovered this story in Justice League of America #144 by Steve Englehart and Dick Dillin, and it turned out the Justice League hid this adventure from the public. The villains battled were the White Martians, who had captured Martian Manhunter.
All six founders were there, although Hal Jordan had yet to become Green Lantern yet. They ended up hiding this battle so they wouldn’t raise the already high martian fears of the public. The Justice League animated series uses this origin as well.
9 Appellaxian Warriors
Since the Justice League refused to acknowledge the battle with the White Martians, their official first teaming was when they battled the Appellaxian Warriors. This story played out in flashback in Justice League of America #9 by Gardner Fox and Mike Sekowsky.
The newest member, Green Arrow, wanted to hear how the team formed, and they told the story of the Appellaxian Warriors attacking Earth, and it took the combined force of all the Justice League founders to stop them. This was also the first time they fought with Green Lantern, although they didn’t know he was the same Hal Jordan who helped them against the White Martians.
8 Starro The Conqueror
The first villain that the Justice League fought in publication continuity came in The Brave and the Bold #28 by Gardner Fox and Mike Sekowsky. This battle put the heroes up against the gigantic sea creature Starro. The monster was able to turn other regular starfish into copies of himself, and it took every member of the Justice League combined to stop the monster. Starro is the same DC villain that showed up in the new The Suicide Squad trailer.
7 Weapons Master
The Justice League of America continued to work as a team in The Brave and the Bold #29 when they battled a villain known as Weapons Master. This villain was a highly intelligent supervillain from the future, arriving from the year 11,960.
This villain found the remains of Wonder Woman’s diary in the future and learned that he was destined to battle the Justice League in the past and lose. However, with his high intellect, he believed knowing what happened could help him better prepare and beat the team this time around.
6 Amazo
In The Brave and the Bold #30, the Justice League battled Amazo. His powers include the ability to replicate the superpowers of any superhero he comes actors. Professor Ivo created the villain. He quickly became one of the most dangerous villains the Justice League faced early in their career together since he could replicate all their powers. The battle began when the Justice League members had their superpowers short out in singles battles and quickly realized a supervillain was responsible.
5 Despero
Justice League of America #1 arrived next as the team finally got their own comic book series. The first villain that the team faced was Despero. This was not the monstrous, super-powerful Despero that he eventually became, but was instead a weak, small man who used mind-control and placing the Justice League members in various battles for his own amusement.
He eventually transported each member of the Justice League to the far reaches of the universe. He thought he won, not realizing they were strong enough to break his control and return to Earth for the final confrontation.
4 Simon Magus, Saturna & The Troll King
Justice League of America #2 saw the team battle magic users for the first time. The storyline started with all the machines on Earth stopping simultaneously, and they soon realized magic caused it and that it was now the dominant force in the universe.
The team realized three evil magicians were pulling the strings in Simon Magus, Saturna, and the Troll King. They came from Magic-Land and wanted to use the science they stole from Earth to overthrow Merlin in their world.
3 Kanjar Ro
Justice League of America #3 saw the team go head-to-head with Kanjar Ro, a space tyrant that wanted to use the Justice League to defeat his enemies. Natural disasters were happening all over Earth, and it is Kanjar Ro who is responsible. He reveals he won’t stop the attacks on Earth unless they help him overcome his enemies. However, when they finish aiding him, he won’t release them, and they have to find a way to stop the powerful alien and save Earth.
2 Doctor Destiny
The fourth issue of Justice League of America featured an alien known as Xandor, but he was not a villain, and it was all a misunderstanding. The next supervillains who appeared came in the next issue when Doctor Destiny gathered together several of the Justice League members individual villains, including Captain Cold, Clock King, Puppet Master, and more. The team then believed Green Arrow betrayed them, but that was all due to Doctor Destiny’s plans.
1 Amos Fortune
Justice League of America #6 saw the team go into battle with a new villain known as Amos Fortune. His power is the ability to manipulate luck to his own ends, and he causes each member of the Justice League to have bad luck and accidents while performing super-feats in front of the public. Eventually, Fortune uses his luck to capture the Justice League, and it is only by his own bad luck that one member was immune from his mechanizations.
